By where you're sleeping

Upgrade to the Rapi:t (¥1,520–1,670) only for guaranteed seats and luggage racks after a red-eye — it saves just 10 minutes. Staying in Namba anyway is the first-timer default.
Umeda, Osaka Station, Shin-Osaka: JR Haruka with the foreign-tourist 1-Way Ticket — ¥1,800 to Osaka Station, ¥1,300 if Tennoji is your stop. The JR Kansai Airport Rapid saves ¥620 and costs 25 extra minutes; fine without luggage, grim with it.
Tennoji or Abeno: Haruka 1-Way at ¥1,300, 30 minutes — the quiet bargain of the table. USJ or the bay: take the Namba or Tennoji route and transfer; there is no direct rail. Groups of 3–4 with luggage: a booked private transfer beats four train tickets on stress if not on yen — see below.
The payment gotcha that matters on night one: Nankai gates accept foreign contactless cards (Visa, Mastercard, Amex, JCB) — JR gates do not. Land, tap, ride to Namba with zero setup; the Haruka requires a ticket first.
